During real-time qPCR, the CT values of the cDNA samples produced with the SensiFast cDNA synthesis Kit lay between 16 and 18. The samples behaved very consistent and results were highly reproducible. The cDNA samples produced with the other kit had CT values between 24 and 28 and were much more scattered.

    Aim: To compare the sensitivity of two commercial faecal DNA extraction kits for the detection and quantification of human hookworms (Ancylostoma ceylanicum, Ancylostoma duodenale and Necator americanus) eggs in faeces.

    Methods: Ten individual human faecal samples positive for hookworm eggs were each extracted with two commercial kits, the Powersoil DNA Isolation Kit (MoBio) and the Isolate Faecal DNA Kit (Bioline) according to manufacturer's instructions. DNA was eluted to 100 ul for each.

    DNA was subjected to multiplex Taq-man based multiplex qPCR (unpublished) targeting the internal transcribed spacer regions of the three human hookworms and an internal control (EHV4).

    Results: Both kits performed equivocally in positively detecting at least one species of hookworm in all ten samples. The Isolate Faecal DNA Kit (Bioline) produced on average 0.5 Ct lower threshold cycles for samples compared to the Powersoil DNA Isolation Kit (MoBio). The time taken to extract 10 samples was significantly less for the Bioline product.

    Conclusion: The Powersoil DNA Isolation Kit (MoBio) and the Isolate Faecal DNA Kit (Bioline) are comparable in terms of sensitivity for detecting and quantifying DNA of hookworms from human faecal samples. The Bioline kit was less labour intensive compared to the Mo Bio kit and is more competitively priced.

    We have been utilizing ISOLATE II RNA Mini Kit from Bioline and have found that the RNA yield, concentrations and purity have been consistently good. From 40,000 liver cancer (mouse and human) cells we are able to get 200-600ng/ul yield of RNA.

    I have been using Bioline products for past 5 years, starting with MyTaqRed Mix and dNTPs. Recently, we started using the SensiFAST cDNA Synthesis Kit and SensiFAST SYBR mix for qRT-PCR. Upon testing with diverse RNA sources, we were really satisfied with the results as the data is quite comparable with competitor SYBR mixes. Quality to price ratio is the winning factor for both the cDNA synthesis kit and SYBR mix. We felt real ease-of-use with the cDNA synthesis kit, which was fast and simple. I recommend the SensiFAST cDNA Synthesis kit and SensiFAST SYBR mix from Bioline.

    I have been using SensiMix SYBR No-ROX Kit to perform RT-qPCR on human and zebrafish cDNA samples with LightCycler 480, and sometimes with ABI ViiA7. I have never had any trouble with the product. The mix has lived up to the promise. I get good melting curves, close Ct values for triplicates, and high PCR efficiency. The melting curve shows very low or no primer dimer. In the manual, there is an example for setting up a 50 µl reaction. I have been using 10 µl reaction, thus making a 500 reactions kit to perform 2500 reactions.

    We have been using MyTaq Red Mix for the past two years in our routine PCR for mice genotyping, cloning/amplicon validation and standard curve preparation for our RT-qPCR. This product is very convenient with gel loading buffer included, and in our hands works as well as competitor products with as low as 5µl of mix per reaction (10µl final volume). The competitive pricing makes it a very attractive alternative for routine PCR.

    I like the Bioline MyFi polymerase, as it can amplify any sequence even if the fragment size is over 3 kb. The polymerase is also useful for cross-species amplification. The figure shows amplification results from meadow fescue using PCR primers designed for perennial ryegrass. All 5 gene sequences were successfully amplified by the cross-species PCR, even though the largest fragment (Gene 4) was over 3 kb in length.

    Our lab has been routinely been using the Bioline SensiFAST Probe Hi-ROX for microRNAs detection in mammalian cells for the past 3 years. Results are highly reproducible between technical replicates and independent experiments. We found that this product showed high sensitivity, low background noise and consistent amplification curves. It performs as well if not better than competitor products. In addition, several thawing/freezing cycles do not influence its activity.
    Given its low cost, SensiFAST Probe Hi-ROX is a really attractive option compared to competitor products. Overall, we are completely satisfied and would recommend it to other labs.

    Working with post-mortem human heart tissue samples for a gene expression project proved to be a challenging task for tissue samples that came from the medical examiner's office. So many factors were working against a viable gene expression profile for these samples such as time of death to autopsy lag, time from autopsy to tissue sample, and whether or not the tissue had been flash frozen. The samples were also never treated with RNA-later so we expected highly degraded RNA from these tissue samples.

    Our only light of hope was the fact that our target amplicon sizes were less than 100 bp which meant that we could still find what we were looking for in spite of highly degraded RNA. Chances were that at least a few intact sequences would amplify. The critical part in our experiment was to be able to yield enough cDNA from whatever bit of RNA we were able to extract from our samples.

    This is where Bioline exceeded all expectations. Working quickly and carefully, I was able to get decent cDNA considering all the factors working against my RNA isolations. The cDNA was enough to be able to carry out a gene expression profile for each of the samples and our project was a success. Bioline proved to be superior in cDNA yield from highly degraded RNA samples.

    I use the MyTaq HS Red Mix all the time for genotyping purposes. This polymerase is so efficient that my program for 35 cycles finishes just under one hour. The mix is easy to use, and the dye in it allows for direct gel loading after the PCR is finished.

    Amazing! Just take a look to the comparison I have made! And I did some others as well: thaw/freeze cycles (4 times without any change), primers specificities (no changes). All the tests were free using small aliquots from Bioline. Only one thing would still stop you? The price is the lowest I've ever had. To me, using my conditions and all the tests I ran, this is the best mix for qPCR!

    We tested the SensiFAST™ SYBR® No-ROX Kit against the LightCycler® 480 SYBR Green I Master on a Roche Lightcycler 480. The cycling conditions we used were optimized for the Roche kit. Despite this, the SensiFAST kit performed significantly better, leasing to earlier amplification from comparable genomic DNA samples (see picture for example). This gives us the increased sensitivity we were seeking. On top of that it is much cheaper than the Roche mix. I highly recommend this product.

    I had been repeating an experiment over and over again because I wasnt getting any amplicon of my target gene after cDNA synthesis and qPCR, thinking my differentiation protocol wasnt working until I tried the SensiFAST cDNA Synthesis Kit and bammmmm, the amplicon was there. I was using more than 50 ng/µl with a different company and wasn't getting any result but with just 10 ng/µl on the SensiFAST cDNA Synthesis Kit, I get to save some RNA for future reference and get the data I am looking for. Are you working with lesser concentration of RNA that you need to be able to replicate your experiments? SensiFAST cDNA Kit is for you then!

    I have used this kit to extract genomic DNA from mice tails (approx. 0.6 cm) for genotyping. Usually a sufficient amount of quality DNA (0.5 to 3 micrograms @ 5-30 ng/µl) from the mice tails was extracted through using this kit- to carry out genotyping. An input of 50 ng for each DNA sample was used for genotyping. This kit comes with very clear, user friendly instructions provided and provides efficient, reliable and cost effective isolation of DNA- overall a great kit for genomic DNA extraction and downstream analysis.

    Our lab has had such trouble from genotyping. We initially started with a simple hot shot method using our own reagents, but the freeze-thaw cycles were degrading the DNA making them unreliable samples. We then began a phenol chloroform procedure to extract the DNA and got great, high quality DNA, but the time versus cost to do this is a problem. Additionally, even with the purest of DNA, some of the samples had to be run on the thermocycler in duplicate again and again to confirm genotype with the Taq we were using. We have faithfully always purchased Bioline’s HyperLadder, and were interested in the MyTaq Extract-PCR Kit when we found out about it at a supplier expo. This kit is absolutely amazing. We went from a two-day purification procedure down to 15 minutes, and the DNA is reliable and survives freeze-thaw cycles. Most importantly, though, the MyTaq is also extremely reliable, and we only have to run samples once to confirm genotyping - and the gels we get are so clean! We have tested all of our mice strains using this kit and all of them work. We are switching to use this permanently from now on! If you have any trouble at all with some of your genotyping, I would give this kit a try.

The MyFi polymerase could amplify complicated short tandem repeat (STR or SSR/microsatellite) sequences better than the other kits. In the figure, the MyFi kit could amplify the STR-A sequence, which the phusion kit could not. The STR-B sequence was amplified with less non-specific amplicons when the MyFi kit was used.

    HypePAGE II prestained protein marker works very well for me, the bands are sharp and contrast well. I have tried it on both Coomassie blue stained gel and regular SDS Page gel, then transfer to membrane. In both cases the bands are sharp and have no smear, and are stable in 4C for at least 3 months.

    We are using the Bioline SensiFAST SYBR No-Rox Mix for our qPCR studies. We have compared the mix to the Roche Lighcycler 480 SYBR Green I Master Mix. With the Bioline SensiFAST mix, our targets regularly appear at lower Cp values and all primers tested work more reliably and consistently (see attached table for two examples of primer efficiencies determined with the Lightcycler and the SensiFast kits). Furthermore, Bioline technical support always answered our queries very promptly and in great detail and the SensiFAST mix costs us less than half the price of the Lightcycler Mix.
